The RNA Folding Problem:   What are the earliest events in RNA folding?

In the absence of suitable quantities of divalent cations, the Tetrahymena ribozyme adopts expanded conformations. Secondary structure is mostly intact, but there is little stable tertiary structure. Following the addition of mM quantities of Mg, the molecule folds up and acquires three dimensional structure that allows it to carry out its biological function.

We are interested in understanding the folding of this molecule, specifically the earliest steps that may share common features with other RNA molecules and perhaps even with proteins!

To achieve this goal, we have developed a unique experimental approach to monitoring the fastest, global conformational changes of RNA following the initiation of tertiary structure formation.

We combine microfabricated tools with brilliant x-ray beams.
